Lung Cancer Update Meets a Stretched Share Price
Iovance Biotherapeutics, Inc. (NASDAQ:IOVA) shares fell sharply in pre-market trading after the company published updated Phase 2 results for lifileucel in previously treated non-squamous metastatic non-small cell lung cancer. The shares had risen 11% to 12% in late Wednesday trading on the announcement of a webcast, so the pre-market decline reversed that overnight gain.
As of an October 14 data cutoff, 17 of 69 patients responded, an objective response rate of 24.6%, in line with the 25.6% reported earlier in 39 patients. The 17 responses include 15 confirmed and two unconfirmed partial responses, and independent radiology review is still ongoing. The study uses a reduced-dose lymphodepletion regimen that has improved safety.
Iovance develops tumour-infiltrating lymphocyte cell therapies. Its lead product, Amtagvi, received FDA accelerated approval in February 2024 as the first TIL therapy for a solid tumour, in previously treated advanced melanoma. The company is headquartered in Pennsylvania, has offices and laboratories in California and Florida, and employs about 975 people.
A Biomarker Strategy Emerges
The more notable finding was the split by tumour genetics. Response was 43.3% (13 of 30) in KRAS wild-type patients and 11.4% (4 of 35) in KRAS-mutant patients, and both complete responses occurred in the wild-type group. Adding HER2 wild-type status and a current or former smoking history narrowed the group to 22 patients, with a 59.1% response rate. That group was 31.9% of the population but produced 76.5% of the responses.
Because the subgroup was identified retrospectively, the company will test it prospectively in POSITILITY, a cohort within the IOV-LUN-202 trial designed with registrational intent, with enrolment starting in November 2026. A frontline trial, OPTILISM, is planned for 2027. In a separate frontline cohort, six of seven similar patients responded to lifileucel plus pembrolizumab, a very small sample. The company estimates the enriched population at about 45% of U.S. non-squamous cases.
Cohorts 1 and 2 are completing enrolment with about 80 patients expected, and a supplemental biologics licence application is planned for 2027. Discussions with the FDA are ongoing.
Valuation Behind the Reaction
Before the drop, Iovance had a market capitalisation of about $6.7 billion and had risen roughly 440% this year, trading near its 52-week high of $15.37 against a low of $1.76. Market data show a price-to-sales multiple near 20 times against about 11 times for the sector, and the consensus price target of about $14.40 sits below recent prices. Trailing EPS is negative $0.73.
Management raised full-year 2026 revenue guidance to $410 million to $420 million. Third-quarter results are due November 5, with consensus revenue near $117 million. The healthcare sector fund was higher pre-market, so the move is stock-specific.
What Investors Will Weigh Next
The data show activity in a hard-to-treat setting, but the headline rate is unchanged and the strongest numbers come from a small retrospective subgroup. Confirmation of the pending responses, FDA feedback on POSITILITY and the November 5 results will shape the next phase.
